Do I need to rotate my fly rod after putting on color preserver so it dries uniformly?

The act of turning it while applying it is usually enough. If you need to rotate it afterwards then you've probably applied far too much. You want to apply enough to completely saturate the threads (they'll turn uniformly dark) but no more.

I turn mine by hand. Color Preserver should tack or set up to the point where it won't run or drip in about 5 minutes. If you apply it a bit too heavily, just watch for sags and turn as needed. Unless you really pour it on there it shouldn't require much in the way of turning or rotating. Certainly not for more than a very few minutes.

Does it take just one coat?

One wet coat is generally enough. If you feel better with two, go ahead. But beyond that you're not really doing anything by adding more.
