Stripper guide placement

Using two stripping guides for a 9' 5wt flyrod, and wondered (based on some previous posts) if the second stripper should be close (4-6") to the first stripper, or should it be moved according to a guide/line distribution test (spaced-out) like the rest of the guides?

No, if you place it according to a static distribution test it won't likely locate in that area. Just try it about 3.5 to 4 inches past your first stripper and see what you think. It may look odd, but if your casting distance improves it suddenly looks a lot better.
