When making the boxes on the wrap on the start of the wrap, does those threads count as one of the passes?

Billy V is the thread master

But i would say yes If you are doing 4 pases and as you put one down -- Now you have 3 pases to do

I never count the initial (layout) threads as the first pass. I consider those as "centering" or "spacing threads and all the remainder as "pattern" threads.

you can count it or don't,doesn't matter much and really is a personal preferance,in the grand scheme of it you're never gonna notice unless it's on a very small diam. blank.remember this is custom rodbuilding, you could do the same exact pattern as i do at the same time side by side and they will be different, just because we're all different.
